Plasma phospholipid transfer protein (PLTP) plays an essential role in HDL metabolism Its role in the transfer of surface remnants from triglyceiide-rich particles to HDL during lipolysis is of importance for the maintenance of HDL levels.It also modulates the size and composition of HDL particles, a function important for the reverse cholesterol transport process. Two forms of PLTP exist in plasm,one catalytically active and the other inactive. A new PLTP ELISA, has been developed whereby pre-treatment of the samples with the detergent SDS leads to the efficient detection of both forms of the protein. The new ELISA is an essential tool when addressing the mechanisms by which PLTP is converted from one form to the other.In addition to its central role in lipoprotein metabolism, a role for PLTP in macrophage cholesterol metabolism has been revealed Differentiated macrophages produce PLTP, and during foam cell formation production of PLTP is markedly increased. As foam cells are a hallmark of early atherosclerotic lesions, further studies on the intra-and extracellular role of macrophage-derived PLTP will advance our knowledge on the function of PLTP in atherogenesis.
