Poly(L-lysine)-poly(ethylene glycol)-folate (PLL-PEG-FOL) conjugate was synthesized to use as a targeting delivery carrier for folate receprot- mediated delivery of proteins or nanoparticles. The synthetic PLL-PEG-FOL conjugate was pre-coated onto the surface of fluorescein isothiocyanate-bovine serum albumin (FITC-BSA) or poly (D,L-lactic-co- glycolic acid) (PLGA) nanoparticles, in an aqueous phase. PLL-PEG-FOL/protein or nanoparticle complexes showed enhanced cellular uptake for KB cells (folate receptor overexpressed cell line) than free protein (nanoparticle) or plain PLL-PEG/protein (nanoparticle) complexes. The presence of an excess amount of free folate in the media inhibited the folate receptor-mediated intracellular delivery of the protein (nanoparticle) by KB cells. The formulation of the intracellular delivery system using the PLL-PEG-FOL conjugate could be also effectively used for folate receptor-mediated endocytosis of other bio-active macromolecules.
