Lipid rafts play important roles in signal transduction and other cellular processes. It has been described that sperm subjected to cryopreservation techniques undergo some physiological modifications that result in similar, but not identical changes in the sperm membrane during capacitation in vitro. This phenomenon is currently known as "cryocapacitation". We examined the formation of raft aggregates in the plasma membrane of macaque spermatozoa incubated in basic media at 4°C for 1 hour and compare these samples with the appropriate controls. In cell membranes, individual rafts are as small as 70-90 nm but the aggregates are much larger domains and can be seen with light microscopy with the aid of DiI-C18 stain that preferentially partitions into ordered lipid domains. Raft aggregates were clearly observed in samples subjected to low temperatures (4°C) similar to those observed in the in vitro capacitated samples. The lipid composition of these raft domains showed a characteristic increase in the content of sphingomyelin and cholesterol compared to the whole plasma membrane analyzed by high-performance thin-layer chromatography. These data suggest that raft aggregation is a dynamic physiological event found in macaque sperm plasma membranes triggered by thermal stress conditions and capacitation.
