Over the past 50 years, the mouse has become the principle investigative animal model of choice for studying cranial suture fusion. In this model, the posterior frontal (PF) suture fuses from day of life 22 through 45, while the sagittal suture remains patent through the life of the animal. Work in this animal model has shown through qualitative and semi quantitative analysis the fusing PF suture and non-fusing sagittal suture to be influenced by TGF-beta1 and FGF2, as well as their receptors. Additionally, some of these genes are known to be responsible for cases of synostosis in humans. Through our previous work, we have shown that real time reverse transcriptase polymerase chain reaction is the most sensitive, reproducible, and quantitative method for analyzing these genes.
