Recombinant glycoprotein therapeutics, such as monoclonal antibodies (mAbs), have revolutionized the treatment of a variety of medical ailments, including cancer, autoimmune and musculoskeletal diseases, and hemostatic disorders. Mass spectrometry (MS) is a rapid, highly informative analytical technique capable of detecting structural changes in peptides and proteins with high selectivity and sensitivity, especially when coupled with reversed-phase high-performance liquid chromatography (RP-HPLC). As such, RP-HPLC/MS is an essential tool for the structural characterization of glycoprotein therapeutic candidates, which are often heterogeneous because of post-translational modifications (PTMs) and differential proteolytic processing. Furthermore, RP-HPLC/MS has emerged as a viable technique to demonstrate product comparability, define cell culture and purification processes, and analyze trace-level protein product impurities.
