As imaging mass spectrometry (MS) and microorganism MS have been developed, sample preparation techniques with raw biological materials (tissues, and microorganisms) have become more important. Matrix-assisted laser desoption/ionization(MALDI) MS has been developed for direct tissue lipids analyses recently.1 However, MALDI MS technique for lipids analyses has two main disadvantages. First, matrix co-crystallization process in MALDI MS doesn't provide spot-to-spot reproducibility through the sample surface. This limits the spatial resolution in imaging MS. Second, it has been reported that phosphatidylcholines (PCs) and sphingomyelins(SMs) suppress other lipids signals in positive-ion MALDI MS mode.2 So, here we present alternative sample preparation technique including graphite3,4 coating. With this technique, direct tissue and cell extract lipids analyses were performed.
