In the current study we used a comparative MS-enabled "shotgun" approach to characterize mitochondrial proteins from young and old rat heart. Our aim is to dissect some of the mechanisms that are potentially linked to the imbalance of the mitochondrial pro- and anti-oxidant systems in the aging heart. A significant challenge for those attempting to use quantitative proteomics is to identify physiologically normal levels of variability and distinguish these from variance that is pathological. One aspect of this challenge is the difficulty involved in processing the copious amounts of data that are needed to describe "normal" variability of results within- and between groups. In this study we used DBParser1 software to condense and compare results from multiple shotgun proteomics experiments. With these results we were able to assess the biological variance in the mitochondrial proteome of young and old myocardium.
