Development of desorption electrospray ionization1 (DESI) has opened up a new area in the field known as "Ambient mass spectrometry~2". Development of DESI was followed by the development of a handful of new ionization methods having one common feature: none of them requires any preparation of condensed phase samples prior to analysis. DESI is differentiated by the fact that it is applicable to large as well as small molecules. The method also allows high throughput analysis and this makes it highly suited to applications to metabolomics and other demanding biological samples. Further unique applications include both in-vivo and ex-vivo mass spectrometric investigations of biological tissues. This presentation discusses DESI in the context of the related ambient ionization methods, especially desorption atmospheric pressure ionization(DAPCI) and direct analysis in real time5 (DART). We also introduce a new method of this general type, Jet Desorption Ionization (JeDI),which employs a high velocity, electrically charged water jet for desorption.
