The proteoglycan decorin is found predominantly in the extracellular matrices of connective tissue (skin, bone, tendon, sclera, cervix and cartilage). Decorin consists of a core protein that is N- and O-glycosylated. The decorin core protein assumes an arch-shaped structure that can form interactions with various extracellular matrix proteins, growth factors and their receptors. The majority of structural studies of decorin have been focused primarily on the glycosaminoglycan (GAG) chain and the core protein. To better understand the roles of post-translational modifications in decorin function, the N-glycosylation patterns of normal decorin needs to be established. Using tandem mass spectrometry, we report a comparison of the expression of N-glycoforms found among decorin from different tissues.
