Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) can be used for the rapid identification of bacterial microorganisms by detection of their unique protein biomarkers. Pathogen identification involves mass spectral pattern recognition of an unknown microorganism to that of a mass spectra of a known microorganism. With the recent availability of microbial genomes, it is also possible to identify microbial protein biomarkers from comparison of observed and genomically predicted protein molecular weights (MW). However, this bioinformatics approach can be affected by the presence of biomarker post-translational modifications (PTMs) which change the MW of the biomarker. We have extracted and identified, by proteomic techniques, several protein biomarkers (and their PTMs) of three strains of Campylobacter jejuni an important foodborne pathogen.
