Lipopolysaccharide (LPS) is a constituent in the outer membrane of gram-negative bacteria and is the endotoxin which would induce the inflammation in the central nervous system (CNS). LPS has used to activate microglia to release pro- or anti-inflammatory cytokines, chemokines, prostaglandins, and nitric oxide. Some of these inflammatory molecules are proteins, which may release into the cerebrospinal fluid (CSF). Some CSF proteins known as brainderived proteins are released by some of the CNS cells (e.g., astrocytes and neurons). Characterization of CSF proteome has the potential merit for the understanding of certain biological events in the CNS. The objective of this study was to discover the biological effects of endotoxin lipopolysaccharide (LPS) induced CNS inflammation of rats by intracerebroventricular administration. In this study, we utilized liquid chromatography tandem mass spectrometry (LCMS/ MS) to investigate the changes of the CSF proteome in rats with CNS inflammation. The 5 days after administration was chosen as the CSF sampling time, which could represent the CNS inflammation. 237 different peptides (186 proteins) were identified by LC-MS/MS. Among 186 proteins, there were a total of 18 proteins with significant differences in LPS group (p <0.05), which contained 16 up-regulated and 2 down-regulated. Three proteins: complement C3, platelet-activating factor acetylhydrolase (PAF-Ahs), and serine/threonine-protein kinase MARK2 were reported to be involved in the inflammatory processes in the CNS. In some literatures, it also reported that Lfucose kinase short variant, calcium activated chloride channel, Shen-dan, and CYP2J9 were presented in the CNS. The proteomic profiling of CSF proteins provide not only information on the abundance of various protein contents after treated with LPS but also comprehension for LPS-induced effects on proteins released in the CNS.
