Nano-Electrospray Ionization Mass Spectrometry (nano-ESI-MS) has become an indispensable technique for the analysis of biologically relevant compounds.1 The ability to couple nano-ESI-MS to powerful separation methods (e.g. HPLC and CE) further increases the usefulness of this technique. While work has been published recently demonstrating the operation of several conventional ESI sources in a parallel, multiplexed arrangement to increase sample throughput, no work has yet been published regarding the multiplexing of two or more nano-ESI sources.2, 3 Consequently, the operational advantages of nano-ESI as compared to conventional ESI have not been fully realized for multiple sprayers; i.e. decreasing sample consumption and improving sensitivity. Furthermore, if the continuous nano-ESI process can be converted into a pulsed mechanism, the favorable characteristics of nano-ESI will be accomplished with the added advantage of even lower sample consumption.4 For this work a novel, dual nano-ESI source has been developed that can be controlled using applied voltages either in pulsed or continuous modes of operation.
