Type 2 diabetes is characterized by insulin resistance in muscle, liver, and fat and by defects in insulin secretion from pancreatic (beta)-cells. Insulin binding to its receptor initiates a complex network of events beginning with the activation of a tyrosine phosphorylation cascade that branches out to affect multiple endpoints. In order to precisely define the tyrosine-phosphoproteome of the insulin signaling pathway we employed stable isotope labeling by amino acids in cell culture (SILAC) in insulin stimulated and unstimulated brown wilde-type adipocytes. To determine more pecisely the roles of the insulin receptor substrate 1 and IRS-2 we compared IGF-1 induced changes of tyrosine phosphorylation in wilde-type, IRS-1, and IRS-2 deficient brown preadipocytes.
