Aspergillus fumigatus is an opportunistic pathogen that causes invasive aspergillosis in immunocompromised hosts, particularly in patients with hematologic malignancies who undergo allogeneic hematopoietic cell transplantation. Innate immunity plays a key role in the defense against Aspergillus fumigatus in healthy individuals. However, recent evidence suggests that cell-mediated adaptative immunity may provide a critical secondary line of defense against the fungi in immunocompromised individuals. As a result, a vaccine that protects immunocompromised patients is being pursuit. Here, we report a proteomics strategy that utilizes a T cell-based bioassay and mass spectrometry to identify vaccine candidates that may protect the immunosuppressed from invasive aspergillosis. CF1, DBA/2 and BALB/c mice were immunized and boosted with either crude Aspergillus fumigatus extracts or with a recombinant fungal protein, rAsp f 3. Afterwards, mice were immunosuppressed with cortisone and challenged with viable A. fumigatus spores. T cells from lymph nodes and spleens of the survivors of the infection were isolated using magnetic beads. The cells were co-cultured together with autologous irradiated antigen-presenting cells and fungal extracts or rAsp f 3 antigens. To identify T cell epitopes, HPLC fractions of trypsin-digest derived peptides of fungal proteins were analyzed by MALDI-Q-TOF, MALDI-ion trap and LC/FTICR-MS and peptide antigenicities were measured in proliferation assays. Concurrently, we tested the HPLC fractions for the presence of antibody-binding epitopes by ELISA.
