To determine the dynamic range of this quantitative assay, we performed the expression levels of proteins which is the exact copy numbers of the mutated Bcr-Abl protein from the CML cell line, BaF3. AQUA strategy is a sensitive method of quantification for Bcr-Abl mutations. Limits of detection (S/N>2) and limits of quantification (S/N>10) for these peptides were found to be 2 femtomoles and 10 femtomoles, respectively. The use of AQUA method may be applicable to monitor dynamic changes of the target proteins before and during therapy with tyrosine kinase inhibotor.
