Redox lipidomics revealed the mechanisms controlling Gpx4-dependent lipid signaling in ferroptosis. Esterified (but not free) oxygenated PUFAs, act as the proximate executioners of ferroptotic death. Lipoxygenase 15 oxidizes PE-(18:0/20:4) and PE-(18:0/22:4) (PE) into ferroptotic signals facilitated by Acsl4-driven esterification of 20:4 into PE.
