Utilizing DIC microscopy in conjunction with fiducial markers significantly improved the accuracy to determine the tissue origin of the species observed in the MALDI IMS in relation to the optical image of the tissue. This approach will assist in identification of these species since the correct region (photoreceptors, inner neuronal retina or RPE) of tissue can now be isolated to improve sensitivity. The localization of these signals will also bring greater understanding of the role RPE65 gene in the visual cycle as molecular constituents originating from the inner neuronal retina not the RPE, have been effected. These data suggest the RPE65 gene affects the visual cycle in a number of varying pathways which expand beyond the retinal pigment epithelium.
