An emergent approach to the detection of nitric oxide (NO) in tissues relies on the use of fluorescence probes that are activated by products of NO autoxidation. Here we explore the performance of the pH-independent, highly selective and sensitive fluorescence NO indicator 8-(3,4-diaminophenyl)-2,6-bis(2-carboxyethyl)-4,4- difluoro-1,3,5,7-tetramethyl-4-bora-3a,4a-diaza-5-indacene (DAMBO-P~H) for the localization of sources of NO in human umbilical vein endothelial cell line, either from endogenous NO synthesis or from chemically released NO from targets of nitrosation/nitrosylation, and explore the specificity of DAMBO-P~H for NO in detail for its application to the biological systems.
