Identification of a Novel Spin Trapped Carbon-centered Radical from COX-catalyzed Peroxidation of Arachidonic Acid:LC/ESR and LC/MS Study

COX-catalyzed peroxidation of arachidonic acid (AA) is a well known free radical reaction. Due to the lack of appropriate methodologies, however, no direct evidence has previously been found for either the formation of COX-mediated free radicals or their associated bioactivities. In order to advance the knowledge of COX-peroxidation, a combination of LC/ESR and LC/MS was used with a spin trap, alpha-[4-pyridyl-l-oxide]-N-rerr-butyl nitrone (POBN), to characterize the carbon-centered radicals formed from COX-catalyzed AA peroxidation in vitro. A total of three types of spin trapped free radicals were characterized, all stemming from PGF_2-type alkoxyl radicals via either (3-scissions or structure rearrangement. A special (3-scission was therefore proposed for the formation of a novel radical which has its electron centered on C=C bond nearby PGF ring. Our results advanced our knowledge of COX-catalyzed lipid peroxidation and will assist our ongoing study of COX-related bioactivities.
