We constructed two forms of catalytically active fused enzyme consisting of yeast CYP51 (lanosterol 14-demethylase) and yeast NADPH- P450 reductase (P450 reductase) to construct a self-sufficient C-C bond cleaving P450.Two fused enzymes named F1 and F2 having different linkers between the CYP51 and the reductase domains were expressed in E. coli cells and purified to homogeneity. The purified F1 and F2 preparations showed spectral properties typical for fused proteins having both P450 and flavoprotein. F1 and F2 showed lanosterol 14-demethylase activity com- parable to that of the reconstituted system with purified CYP51 and P450 reductase. The rates of the enzymatic reduction of the P450 in F1 and F2 were somewhat faster than that observed on the reconstituted system.
