Prokaryotes use alternate sigma factors of RNA polymerase for transcriptional selectivity in response to different stresses. rpoS encodes the stationary phase sigma factor, (sigma)~(S), which is required for proper expression of about 100 stress-response genes. When nutrients are abundant, SprE binds RpoS and delivers it to ClpXP for degradation, but when nutrients are absent this process is inhibited. Overproduction of SprE causes a toxicity phenotype, seen as a severe growth defect. Based on the known function of SprE, it is not inherently obvious of how this protein could be toxic. Null mutations in pcnB [encoding poly(A) polymerase I (PAP I)] and hfq were found to suppress the toxicity phenotype. PAP I in conjunction with the RNA chaperone Hfq adds poly(A) tails onto the 3' ends of mRNAs which target them for destruction. Here we use biochemical assays to demonstrate that SprE has a second function involved in the PAP I polyadenylation pathway and hence control of mRNA stability. We show that PAP I interacts with the degradosome in both growth phases, and this interaction is dependent upon SprE during stationary phase.
