50percent DMF was confirmed to provide sufficient solubility for each MALDI matrix up to 0.2M concentration. The measured and calculated parameters used to compare these matrices are good tools to rank and categorize matrix performances in MALDI-TOF MS measurements. The evaluation of matrices through crystal morphology, UV absorbance, extinction coefficient and hydrophobicity (logP) does not provide enough data to determine which is the best matrix. The determining factors seem to be the size of the molecule and its ability to stack over each other in the crystals. As shown by the MALDI-TOF MS data, the use of 3-HPA provided the highest S/N ratio and mass resolution. More studies are required to understand and correlate each parameter to the MALDI-TOF MS data.
