1. BRD3 is overall a very structured protein with the majority of the flexibility occurring in the ZA and BC loops; 2. The ZA loop has the most change in dynamics upon JQ1 binding; 3. In BRD3(1) binding to JQ1 the ZA loop shows more protecti on from deuterium exchange at earlier time points while in BRD3(2) binding to JQ1 there is more protection at later time points suggesting differ ences in ZA loop dynamics upon binding; 4. This study highlights the importance the ZA loop may play in ligand binding.
