We performed SILAC-based protein correlation profiling on mitochondrial membrane vesicles. Our data allowed us not only to assign proteins to the outer or inner mitochondrial membrane, but also to discriminate mitochondrial membrane proteins from contaminating organelles such as the endoplasmatic reticulum or vacuole. We identify proteins which were not previously known to reside in or to be associated with mitochondrial membranes. We further discovered MICOS, a multisubunit complex required for the formation of contact sites between both mitochondrial membranes.
