Francisella tularensis (Ft), a type of gram-negative bac terium, is best known as the cause of tularemia, an infectious disease in humans with low infectious dose and high mortality rate. Lipopolysaccharide (LPS) has been reported as a protective antigen (1). Three types of saccharides in Ft LPS that contain O -antigen (OAg) were described in our recent report, two consisting of tetrasa ccharide repeats attached to one of two core oligosaccharide variants, and one consisting of tetrasaccharide repeats without core (2). Since all three types of OAg-containing saccharides have potential to generate an immune response toward Ft, preparative chromatographic separation of glycans of defined length and composition is necessary to map epitopes targeted by protective anti-Ft LPS antibodies towards development of vaccines and immunotherapeutics for tularemia. This report details the successful preparation of pure LPS saccharides by a two dimensional chromatography method, as demonstrated using mass spectrometric analysis of the fractions.
