A quantitative capillary HPLC-ESI -MS/MS based methodology has been developed to accurately quantify CPA-induced G-NOR-OH monoadducts and G-NOR-G cross-links in HapMap human cell lines and peripheral blood lymphocytes. The new methodology was employed to investigate the formation and repair of CPA-induced DNA adducts in PBLs isolated from blood of ten healthy volunteers and in five human HapMap cell lines. Our results reveal significant inter-individual differences in the kinetics of G-NOR-G adduct formation and repair. Future studies will investigate the usefulness of DNA damage assessment in PBLs treated with reactive metabolites ex vivo as a predictive model of in vivo efficacy and toxicity in patients receiving CPA and other DNA alkylating agents.
