Distinguishing astrocytic from oligodendroglial differentiation in diffuse gliomas is a diagnostic challenge in surgical neuropathology. New markers are constantly being sought to aid in glioma classification [1]. Glial fibrillary acid protein (GFAP) is a reliable immunoperoxidase marker of astrocytic differentiation, but also stains minigemistocytes and gliofibrillary oligodendrocytes.CD2 is a T-lymphocyte cell-adhesion molecule that mediates adhesion of T cells to antigen-presenting cells [2,3]. This marker is used in the investigation of T-cell neoplasms. To date, CD2 expression has not been reported in glial or neuronal cells.A new antibody raised against CD2, code: SG-CD2 (D2) was incidentally found to stain a subset of astrocytic cells in central nervous system (CNS) neoplasms. Such a pattern of immunostaining is not seen with a commercially available monoclonal antibody against CD2 (Novocastra, UK). This study aimed to investigate the staining pattern of D2 in a variety of neurosurgical specimens. Of particular interest was whether D2 was useful in distinguishing astrocytomas from other gliomas, in particular oligodendroglioma.
