In this study, we employed cDNA microarray analysis to identify genes up-regurated by Procain (PCA) treatment in gastric cancer cell line and compared with genes up-regurated by DAC treatment. We surveyed up-regulated genes to discover aberrantly methylated gene and determined the methylation status of 5'CpG islands by bisufite DNA sequencing.As a result of classification as for function, genes related to "cell cycle" and "apoptosis" were up-regulated by both PCA and by DAC treatment. However, there was a significant difference in up-regulated genes between PCA and DAC treatment, suggesting difference of the growth inhibitory mechanism. Using bisufite DNA sequencing, a gene related to apoptosis, diva was found to be transcriptionally repressed by promoter hypermethylation and this gene might be related to gastric carcinogenesis.
