Stachys, one of the largest genera in the flowering family Lamiaceae, have been used as ornamental plants, edible food sources and as therapeutic agents in traditional medicine. Stachys natalensis Hochst. is a perennial, erect herb with white flowers and faintly aromatic leaves which are densely covered with hairs on both foliar surfaces. Very little information is available on the structure of the secretory apparatus and nature of the secretions (which may be of therapeutic value) found on the foliar surface of S. natalensis. This study investigated the micro-morphology of the abaxial and adaxial leaf surface structures of S. natalensis utilizing stereo microscopy and scanning electron microscopy (SEM). Leaf material, across three developmental stages (emergent, young and mature), were conventionally freeze-dried, chemically fixed as well as cryo-preserved. The processed samples were mounted onto stubs, sputter-coated with gold and platinum for conventional and cryo- electron microscopy respectively, and viewed using the Zeiss Ultra Plus Field Emission microscope and SmartSEM image software. The origin, nature and chemical composition of the secretions were also determined using various histochemical staining procedures. Fresh and dewaxed semi-thin leaf sections were stained for the detection and localization of chemical compounds and viewed using the Nikon Eclipse 80i light microscope. Images were captured using the NIS-D elements software.
