Because of growing interest in the biological properties ofD-allose, its "F-labeled analogue, [18F]2-fluoro-2-deoxy-13-D-allose ([~(18)F]2-FDβA), was synthesized to probe its biologicalactivity. A fast and efficient two-step, regio-, and stereoselectivesynthesis of [~(18)F]2-FDβA was achieved by electrophilicfluorination of 3,4,6-tri-O-acetyl-D-glucal in anhydrous HF using[~(18)F]F2and [~(CH3COOF. The fluorocarbohydrate wascharacterized by multi-NMR spectroscopy and mass spectrometry.Small animal PET imaging was used to study the in vivo behaviorof [~(18)F]2-FDβA. Preliminary studies using Polynoma Middle Tmice showed that [~(18)F]2-FDβA is a promising radiotracer for thedetection of breast tumors.
