Biomimetic Interfaces for Integrated Biosensor Arrays

Protein-based electrochemical biosensors are well suited for direct detection of a variety of chemical and biological analytes. Biomimetic interfaces consisting of synthetic lipid bilayers allow proteins to be immobilized in an active conformation on an electrode. The electrode allows transduction of the protein's activity into an electrical signal, thus yielding a biosensor. This paper describes ongoing research to integrate biomimetic interfaces with a fully scalable, three-electrode system on a silicon chip. The resulting integrated biosensor could be readily miniaturized to produce high-density, multi-analyte arrays. Methods for depositing lipid bilayers and proteins on electrode surfaces are discussed in this paper, along with characterization of the resulting biomimetic interfaces by a variety of analytical techniques.
