The cryopreservation effect is usually measured by percentage pf motile spermatozoa after freezing/thawing procedure but these is not always reflected in fertilization success. The best method for verification of efficiency of cryopreservation biotechniques is hatching rate determination after fertilization with freezing sperm (Glogowski, et al. 2002). It has been estimated that dimethylsulphoxide (DMSO) commonly used cryoprotectant is hydroxyl-free radical producer (Miller and Cornwell, 1978) and during cryopreservation harms the DNA which results in lethality during the embryo development. On the other hand, Horvath et al. (2000) proved that methanol (MeOH) used as cryoprotectant affects motility worse than DMSO, but results in good biological effect shown in hatching. The aim of this experiment was optimalization of cryopreservation biotechnique of Atlantic salmon sperm by testing eight variants of cryoconservation. The testing factors were: cryoprotectants (DMSO and MeOH), extenders (glucose and sucrose) and technique of freezing (pellets and straws).
