Three types of wheat ra-gliadins were identified previously, based on N-terminal sequences. Types beginning ARQ or RQ were separated from types beginning KEL by acid PAGE and ion exchange chromatography and referred to as co-2 and co-1 types, respectively1'2. N-terminal sequences of ARQ and KEL types were similar except for the first 8 amino acids of the ARQ type. ARQ and KEL types were associated with chromosomes 1A and ID of bread wheat. A third type beginning SRL was associated with chromosome IB, and referred to as (0-5 type. Since it was not clear how to distinguish between the co-gliadins from chromosomes 1A and ID based on protein or gene sequence data, co-gliadins were purified from T. urartu, the probable donor of the A-genome', and compared with 1A co-gliadins that were purified from the bread wheats Buile86 and Chinese Spring nullisomic ID tctrasomic 1A (CS N1DT1A). N-terminal sequences and masses of the purified co-gliadins suggested that the KEL types (co-l) are derived from ARQ types (co-2) by cleavage between an asparagine and a lysine. Data presented elsewhere indicate that 1A and ID co-gliadins can be separated by HPLC and that 1A co-gliadins are generally somewhat smaller that ID co-gliadins but are similar in terms of N-terminal sequence.
