Hypothetical Differential Processing of Low Molecular Weight Glutenin Subunits Met- and Ser-types at Their N-Terminal End

The gluten polymeric fraction is composed by high and low molecular weight glutenin subunits (HMW-GS and LMW-GS, respectively). Typical LMW-GS are classified into two main groups, namely LMW-s and LMW-m types, whose first N-terminal amino acids in themature polypeptides are Ser and Met, respectively, along with a third minor group, LMW-i, storting with He. It has been hypothesized that the presence of the Asn residue instead of a Tiff residue, as it occurs in the supposed LMW-m type sequences, coulddetermine a differential processing at the N-terminal end of LMW-s type sequences, that might generate the cleavage of the peptide MEN by an asparaginyl endoprotease. In order to verify this hypothesis, we have expressed two LMW-GS in Nicotiana benthamiana by using an episomal vector based on Potato Virus X (PVX). The two LMW-GS expressed are represented by two wild types LMW-m and LMW-s genes, along with the mutated forms at position 23. Preliminary results give indications of occurrence of such a differential gene processing at post translational level, at least in the LMW-m type.
