We investigated peripheral blood lymphocyte subsets and Thl/Th2 cytokine serum levels before and in the course of six months following successful autologous hematopoietic stem cell transplantation (aHSCT). The highly cytotoxic CD 160+ NK subset progressively recovered from M2 to M6. Reconstituted T cells comprised mostly effector memory (EM) and effector (E) CD8+ subsets, and CD4+ T were antigen-experienced Th1-like central-memory (CM) cells. While IL-10 levels before aHSCT were either too low (mean 3.8 pg/ml) or too high (mean 10.5 pg/ml), (MW p< 0.001), they were modulated within the reference interval (3-11 pg/ml) at M6. Moreover, IL-10 concentrations at M2 and M6 correlated positively with IFN-gamma (R=0.36), the proportion and absolute count (AC) of NK cells (R=0.4), and - negatively with the proportion of activated (CD57+CD28+) CD8 T cells (R= - 0.76). Thus, IL-10 may regulate post-aHSCT reconstitution by modulating Th1 immune responses, and eventual GVHD-like effects.
