Isolation and characterization of cellulase producing bacteria from pruning tree compost and soil

Twenty-seven strains of moderately thermophilic bacteria, which degraded microcrystalline cellulose or carboxymethylcellulose (CMC), were isolated from pruning tree compost and soil. The 16S rDNA sequence analysis indicated that these isolates were classified into four groups in species level. The two groups of the four showed less than 95% similarity with any published 16S rDNA sequences and were related to the sequences of Paenibacillus spp. This result indicated that isolates of these two groups are novel species of the genus Paenibacillus or possibly novel genus. Strains SCA3a and strain TC22-2b were selected as representatives of the strains of the two groups and were characterized physiologically and chemotaxonomically. The optimum temperature and pH for growth of both SCA3a and TC22-2b were around 55°C and 7-8, respectively. This optimum temperature was at least 20°C higher than that of closly related published Paenibacillus species. Also the major fatty acid compositions of strain SCA3a and TC22-2b were not similar to the related species.
