The persistence of faecal Bacteroides species after release into differing environmental waters and sediments is not well understood. This 18 day study aimed to determine the survival (using culture-based method) and PCR-based marker recovery of human- and dog-sourced Bacteroides from river and seawater laboratory microcosms containing muddy sediment. The survival of Bacteroides in the water declined very sharply from 1.2×10~4 cfu/100 ml to near zero between day 6 and day 14; likewise for those in the sediment a rapid decline was observed from 8.0× 10~3 cfu/gram to near zero between day 8 and day 16. Generic, human and dog Bacteroides PCR primers were used to evaluate the persistence of faecal Bacteroides markers in these controlled conditions and the rates of decline were similar to culture-based recovery. The similarity in persistence of these Bacteroides 16S rRNA genetic markers in environmental water and sediment suggests that viable but non-culturable (VBNC) Bacteroides do not persist in the natural environment for long and therefore that 16S rRNA markers can thus be of value as faecal indicators of bathing water pollution and in source tracking.
