This survey aimed at investigating the arsenic (As) speciation and the responses of the enzymatic antioxidant system in Pityrogramma calomelanos under As stress. Ferns at 4-5 frond stage were grown in half-strength Hoagland's solution containing 0 or 1 mM of As. After 21 days, the As concentrations in pinnules, raquis + stipe, and roots were around of 3108, 275 and 283 mg As kg~(-1), respectively. As was present mainly as arsenite (pinnules, 75%) and arsenate (raquis + stipe 74%, roots 92%). Exposure to As increases the activities of APX and POX in pinnules, and SOD and CAT in roots. Induction of antioxidant enzymes is essential to quench reactive oxygen species to a low level, thus, enhancing continuous As accumulation. Reduction of As is also important to increase the translocation and sequestration in shoot. These results help to understand P. calomelanos As hyperaccumulation.
