In this paper, the 80% ethanol extract from the leaves of Dendrocalamus hamiltonii were divided into four fractions of petroleum ether extract, chloroform extract, ethyl acetate extract and water extract. These were used to determine the flavone content using rutin standards and colorimetry of NaNO_2- Al (NO_3)_3 at 510 nm. The regression equation of rutin standard working curve is established as Y=0.1045X+0.0179 (R_2=0.9986). Free radical scavenging activities of chloroform extract and ethyl acetate extract were measured with the DPPH- method. The experimental results indicated that the flavone contents of petroleum ether extract, chloroform extract, ethyl acetate extract and water extract are 5.60%, 15.38%, 9.16% and 1.42% respectively. The DPPH- IC50 scavenging value of chloroform extract and ethyl acetate extract are 0.21 mg/mL and 0.13 mg/mL respectively. The results demonstrate that the flavone content of the chloroform extract and ethyl acetate extract is higher than that of petroleum ether extract and water extract; and the DPPH- free radical scavenging activities of ethyl acetate extract is greater than that of the chloroform extract.
