In multiplex real-time PCR specific parts of the genome from different microbial target species are multiplied, quantified and detected simultaneously. In assays developed for diagnosing bovine IMI, PCR test panels include predesigned oligos for mostcommon mastitis causing pathogens (Koskinen et al., 2009). Objectivity, short turn-around times and higher sensitivity have been listed as advantages of PCR over the conventional bacterial culture (BC). PCR amplifies DNA also from dead and growth-inhibited pathogens. BC negative samples often yield positive PCR results (Taponen et al, 2009). This is considered as an advantage in some contexts and as a concern in others. Accepted guidelines for interpretation of PCR results are lacking. The aim of this short review was to highlight factors related to sampling technique and timing of milk sampling, which affect PCR results.
