To isolate antigen specific antibodies is critical for a variety of diagnostic, therapeutic, and research purposes. For efficient isolation of antibodies against a target antigen at ease with bypassing animal immunization, we have constructed three kinds of antibody libraries, synthetic, semi-synthetic, and human na?ve immune libraries, composed of a huge number of individual transformants (size: > 10[10-11]) for the display of single chain variable fragment (scFv) antibody variants on the minor coat protein of bacteriophages. To improve the quality of the antibody libraries, both growth based selection and FACS based selection were employed and the utility of the constructed libraries were validated by screening against three model antigens.
