Objective: To investigate the aberrant methylation of P16 and adenomatouspolyposis coli (APC) gene in peripheral blood with liver cancer patients andevaluate their clinical significance. Methods: P16 and APC gene aberrantmethylation in liver cancer tumours, pericarcinous tissues, and their correspondingperipheral blood were detected by using real-time quantitative polymerase chainreaction (Real-time PCR) in 86 hepatocellular carcinoma (HCC) patients. Results:The methylation rate of P16 and APC gene in liver cancer tumor tissues wassignificantly higher than that in pericarcinous tissues and their correspondingperipheral blood (p < 0.05). The coincidence rate of methylation of P16 and APCgene in cancer tumor tissues and peripheral blood was 65.4% and 43.3%,respectively. The P16 gene methylation rate of peripheral blood in HBsAg positivegroup was higher than that in HBsAg negative group (p < 0.05). Conclusion: Thedetection of P16 gene methylation in peripheral blood may be used as an index inearly diagnosis and surveillance for liver cancer patients.
