This study aims to investigate deltamethrin resistance in Culex pipiens pallens and its association with knockdown resistance (kdr) mutation as well as to explore the establishment of a mosquito-resistance testing method. Using specific primers through polymerase chain reaction (PCR), kdr gene sequences isolated from wild Cx. pipiens pallens mosquitoes were sequenced. Linear regression analysis was used to determine the relationship between the mutation and deltamethrin resistance. Two alternative fair-like mutations, L1014F and L1014S, were positively related to the deltamethrin-resistant phenotype. A novel mutation, Thymine Cytosine Thymine (TCT), was identified for the first time in the Voltage Gated Sodium Channels (VGSC) of Cx. pipiens pallens. Three fair-like mutations in Cx. pipiens pallens were positively related to the deltamethrin-resistant phenotype.
