Determining viability of porcine mycoplasma cultures using a redox indicator

Mycoplasma hyopneumoniae (Mhp), Mycoplasma hyorhinis (Mhr), and Mycoplasma hyosynoviae (Mhs) have caused a big economic loss in the pig industry worldwide. In order to prevent mycoplasma infection, pure cultures were submitted from university veterinary diagnostic laboratories to federal licensed biological companies for autogenous vaccine production. PCR assays are used to identify mycoplasma species. However, PCR results cannot tell the difference between viable and dead cultures. In order to determine viability of the submitted cultures, a non-toxic redox (oxidation-reduction) indicator (alamarBlue) was used. The alamarBlue (life technologies) assay is designed to monitor the reducing environment of proliferating mycoplasma culture.1 If the tested culture is alive, innate metabolic activity of mycoplasma culture will make alamarBlue in reduced form (pink in color). If the tested culture is dead, alamarBlue will be in the oxidized form (blue in color).
