Certain metal ions induce hydrolytic degradation of RNA, and in some RNA molecules this process is exceptionally efficient and specific. The best known example, yeast tRNA~(Phe), undergoes specific fragmentation in the D-loop in the presence of Pb~(2+) [1-3] and other ions: Eu~(3+)[4,5], Mn~(2+)[6] and Mg~(2+)[7,8]. It has been shown, based on X-ray analysis of yeast tRNA~(Phe) crystals, that a Pb~(2+) ion induces the reaction acting from its tight metal ion-binding pocket [9,10]. This gave rise to an experimental approach that uses Pb~(2+) and other ions to localize some metal ion binding sites as well as to probe the structure of RNA molecules in the vicinity of the bound ions.
