Abstract: Rat adrenal glands fluoresce intensely after systemicapplication of meso-tetra(hydroxyphenyl)chlorin(mTHPC). We investigated which parts of the adrenalgland accumulate mTHPC. Furthermore we examined thetime course of adrenal mTHPC-accumulation. Ten maleWistar rats each were given 0.5 or 0.7 mg mTHPCkg$+$MIN@1$/ iv. Each two animals were perfused withnormal saline and Zamboni fixative 6, 12, 24, 48 and 72hours after photosensitization. Untreated animalsserved as controls. Fluorescence was quantified on 20micrometer frozen sections with CCD-camera andappropriate software. Immunohistochemistry identifiedspecific cell types with antibodies tosteroid-synthesizing enzymes. The cortex exhibited anintense fluorescence, with weaker fluorescence ofcorticocytes in the zona glomerulosa compared to theother zones. Besides intensely fluorescing singly lyingscattered cells, the medulla showed a faintmTHPC-induced fluorescence. Immunohistochemistryrevealed that intramedullary cells with intensefluorescence were corticocytes, showing a positivereaction to the 21-$beta@-hydroxylase antibody. Peakaccumulation of mTHPC was always observed after 24hours. Our results indicate for the first time thatonly steroid synthesizing cells of the adrenal glandexhibit an intense photosensitizer-inducedfluorescence. Thus mTHPC-application is anuncomplicated method to identify steroid-synthesizingcells, possibly also in other organs. !15
