The aim of this study was to investigate the longterm cultivar characteristics (fruiting body yield and sizes) in order to provide spawns for shiitake mushroom growers. The 6F1, 7F2, and 9F3 Shiitake mushroom cultivars were employed. Malt saccharified extract agar (MSA), MSA plus minerals (MSMA), potato dextrose agar (PDA) plus minerals plus corn steep liquor (PDMCA), and PDMCA plus Japanese oak sawdust (PDMCSA) were used as preservation media. Sawdust box culture was adopted (1.2 kg/box; Japanese oak sawdust plus wheat bran) for fruiting body cultivation. The fruiting body yield in the most mated dikaryotic mycelia with the same parental cultivar and stocked on MSA medium decreased by 50% in 1-2 years. During the spawn manufacturing process, repeated subculture on agar media also caused a decrease in yield. The use of different agar media and/or subculture frequency led to unevenness in yield. The fruiting body yield of the cultivar preserved on MSMA for 1 year was stable. Each cultivar required optimum preservation media for sustaining characteristics. Moreover, the parental cultivar and its F1 hybrids also required optimum preservation media. Isolating the fruiting body tissue is effective for sustaining cultivar characteristics. Thus, it is necessary to determine whether hybrids sustain their characteristics after 2 years of subculture, to choose the optimum preservation media for each cultivar, and to isolate cultivated fruiting body tissue.
