Statistical Optimization of Process Variables for the Production of Mycelial Biomass and Polysaccharides from Lentinus edodes

Physiologically-active polysaccharides are generally obtained from commercially cultivated mushroom fruiting bodies, which have a long cropping cycle. However, growing mushroom mycelia in a defined medium, by submerged fermentation is a rapid and alternative method to obtain fungal biomass of consistent quality for the extraction of polysaccharides (intracellular and extracellular). Response Surface Methodology (RSM) was used for statistically optimization of the process variables such as pH (4-8), temperature (15-351), incubation period (10-30 days) and agitation rate (46 -54 rpm) in liquid medium for maximum biomass production of 7 strains of L. edodes. Out of these, one strain LeI produced maximum biomass and polysaccharide. Central Composite Rotatable Design was applied to design experiments to evaluate the interactive effects of process variables using Design Expert, Trial version 8.0, Stat-Ease INC., Minneapolis, MN statistical software. The second order polynomial coefficients were calculated using the software and responses of the RSM were plotted on 3-D plots. The optimum operating conditions obtained by RSM were pH 5.0, temperature 21℃, incubation period 22 days and agitation rate 52. 0 rpm for maximum biomass (7.93 mg/mL) and polysaccharide (1. 67 mg/mL) production in LeI strain. These optimal conditions are being used for large scale fermentation for simultaneous production of biomass and polysaccharides.
