While the importance of heat shock proteins (HSPs) in the heat resistance of organisms is becoming widely accepted, the expression of HSPs in thermal tolerance may not be the same among different tissues. In this study, the common carp Cyprinus carpio was utilized to determine tissue-specificity of HSP70 proteins expression in vivo 24 h acute heat shock exposure. Upper thermal tolerance was firstly determined by a standard method to obtain the temperatures of acute heat shock. In exposure experiments, fish were kept at control for 10 ℃, or subjected to an acute heat shock for 24 h at 20 ℃, 24 ℃ and 28 ℃, or subjected to 20 ℃ and 28 ℃ heat shock and then allowed to recover, at control temperature for 24 h. Levels of HSP70 in the brain, kidney, gill, heart and muscle were determined by Western blots of one dimensional SDS-PAGE. Unlike heart, kidney, brain and gill appearing as a single band, the selected antibody recognized two protein bands in muscle. Prominent induction of HSP70 was detected in heart, gill and kidney following a 10–18 ℃ increase in temperature, while obvious decreased levels of HSP70 in gill was observed at 28 ℃ heat shock. These results suggest heat stress caused variations in the synthesis of HSP70 in different tissues of C. Carpio.
