Triple negative breast cancer (TNBC) behaves more aggressively and has a poorer survival rate compared to other types of breast cancers. Dasatinib is an orally active small molecule drug that targets kinases. Most triple negative breast cancer cell lines are sensitive to the in vitro anti-proliferative activity of dasatinib, but some are resistant. We sought to use high throughput screening to identify compounds that enhance the activity of dasatinib towards a dasatinib-resistant TNBC cell line, MDA-MB-468, but which have no effect by itself. We optimized a cell viability assay in 96-and 384-well plates for high throughput screening. We screened the Prestwick library of FDA-approved drugs and identified one unconfirmed active, cantharidin. A less potent analog of cantharidin did not show differential activity with and without dasatinib. During the course of the screen, the natural plant product parthenolide was identified as having activity by itself against this cell line.
